How does C-Stem™ achieve almost 300-fold hiPSC expansion within a week?

Following a recent peer-reviewed publication describing a 275-fold amplification of human induced pluripotent stem cells (hiPSC) in a 10L bioreactors in 6.59 days with the C-Stem™ technology, Maxime Feyeux, PhD, co-founder and Chief Scientific Officer of TreeFrog Therapeutics, explains how such exponential growth can be obtained.

It’s a question that we often have: how do we have a 300 fold-ish expansion over a week of pluripotent stem cells in C-Stem™?

That’s not really a surprise to me. Because, actually when you look at what happens in vivo, you have an organized, protected environment in which the cells (the primed pluripotent stem cells) go from about 10 cells to thousands of cells actually. So that expansion is basically what you expect to have.

The question that we could ask in reverse is: why don’t we get that with normal culture systems, like classical culture systems? In 2D and in aggregates, the cells are not really protected. They don’t recapitulate the proper shape. And actually I think the lack of reproducing a proper topology, a proper protected topology, is the reason why we don’t get that expansion.
